Before UFC light heavyweight Jon Jones turned down a fight with Chael Sonnen, the two engaged in a war of words. Jones had just beaten a former champion by submission, but the mere sight of Evans ruined what should have been a jubilant evening for him.

When Jones apologized for getting behind the wheel of a car while drunk, he lashed out again at his "haters" who tried to "kick him while he was down." After White announced the cancellation of UFC 151 and lashed out against Jones and his camp, Jones actually unfollowed his boss on Twitter.

For a millionaire professional athlete, Jones has a thin skin. He often appears more concerned with what his detractors think than his supporters.

It's an odd stance for someone of his level to take, because part of the job of being a famous athlete is dealing with people who don't like you. Abuse allegations mount against flagship Afghan police force

CHAR DARAH, Afghanistan (Reuters) - Fahima had just arrived home from school when members of the Afghan Local Police (ALP), a U.S.-trained militia charged with making Afghans in Taliban strongholds feel more secure, started hammering on the front door searching for her father.

They elbowed it open and, frustrated at not finding him, started beating her younger brother, prompting 17-year-old Fahima to intervene. One of the men turned and shot her dead.

"She was in her first days as an eleventh grade student," said Fahima's father, Khuja, who believes the killing was score settling over an old land dispute.

"Offenders are still serving as local policemen and they are free. Police say the killer has escaped but he's walking in public with his gun and no one is able to catch him."

The ALP was set up in 2010 in villages where the national force is weak, a flagship project of U.S. General David Petraeus, who stepped down as commander of foreign forces in Afghanistan in 2011.

The government began recruiting everyone from farmers to shopkeepers for the militia, hoping to take the edge away from the Taliban in their rural bastions.

American officials have hailed the ALP as an effective homegrown force which has restricted the ability of the Taliban to move in the countryside.

In northern Kunduz province's Char Darah district, a Taliban stronghold until recently, people credit the ALP for making it safer to travel and send children to school against frequent insurgent opposition to education, especially for girls.

"The Taliban here were demanding money from local people, beating them if they refused. Now we don't let them do it," said Gul Ahmad, an ALP commander in Sarak Bala village.

But security gains made by the now 20,000-strong militia are often overshadowed by mounting accusations of abuses, including rape and murder.

Human rights groups say ALP members sometimes act like warlords, demanding bribes, skimming contracts and committing the kind of atrocities that rattled Afghanistan in a civil war that killed 50,000 people before the Taliban took over in 1996.

Afghans already have enough to worry about. Many fear the United States and other Western allies will abandon Afghanistan after 2014, when most NATO combat troops will have gone, leaving them at the mercy of the Taliban. There is widespread talk of another civil war.

The ALP was supposed to ease public anxiety, not fuel it.

UNIFORMS, A SALARY, BUT LITTLE DISCIPLINE

Duties range from manning checkpoints and running patrols to providing security forces with intelligence on insurgents. Each member gets a monthly salary and food worth about $180 and are issued brown uniforms and an AK-47 rifle.

Some acquire heavier weapons like machineguns or rocket-propelled grenades on their own and prefer the traditional flowing shirt and baggy trousers to mix in with the population in farming villages with mudbrick homes.

Many complain they are underpaid and have to borrow or steal from the poor locals they are meant to protect.

"My father works as a farmer and I have to help him live. If I don't get enough money then I'll have an eye on other local people's pockets," said Lutfullah, 28.

Their pasts often don't inspire confidence either. Rights groups say some were former Taliban fighters or members of militias that wreaked havoc in Afghanistan for decades. There are reports of the ALP joining the Taliban.

"Some of them are guilty of repeated killings," said Hussain Ali Moin, coordinator for the Afghanistan's Independent Human Rights Commission.

More than 100 ALP members have been jailed for crimes including murder, bombings, rapes, beatings and robbery, according to chief military prosecutor Mohammad Rahim Hanifi.

In one of the most high-profile cases, an ALP commander and four of his men entered a house in Kunduz province, assaulted a family and abducted their 18-year-old daughter, Lal Bibi, in May.

She told her family she was chained to a wall and repeatedly raped before being brought home a week later.

"She says if she does not get justice she will set herself on fire," her 56-year-old father, Hajji Rustam, told Reuters.

The trauma was so severe, it made him long for the days when rapists were publicly stoned to death or flogged under Taliban rule.

"The Taliban were better than the ALP," he said. "At least they respected our honor. They opposed only women's activities in public, but these people assault us in our homes."

STRICTER VETTING

The problems may multiply, with plans to boost the force to 30,000 and make it operational over most of the country.

Some of the attacks allegedly committed by the ALP also seem to be motivated by sectarian rivalries, which could complicate efforts to tame the force.

In southern Uruzgan province, an ALP commander belonging to the Hazara minority ethnic group in late July gunned down 15 Pashtun civilians in Khas Uruzgan, a day after the Pashtun Taliban killed two of his friends, officials said.

"Commander Abdul Hakim Shujahi took nine villagers out of their houses and took them to the Matakzai area of the village and killed them with stones and gunshots," said Mohammad Waris Faizi, who heads the Independent Human Rights Commission investigation office in the province.

"Then he and his people arrested six villagers from the Khak Afghan area and killed them too," Faizi said.

Interior Ministry spokesman Sediq Seddiqi denied troubles were widespread and said the ALP was making serious sacrifices for Afghanistan, accounting for more than half of all casualties among anti-insurgent forces.

"They are very useful, to be honest, and very effective. They are inflicting big losses on the Taliban," he said.

In order to improve the ALP's image, authorities say they are stepping up vetting beyond word of mouth from local leaders, governors, intelligence and police chiefs, and turning to biometric data.

"If there are any criminals, we can track them and we can find them," Seddiqi said.

But those efforts have not calmed Afghans.

"The Taliban and ALP are active during the night," said villager Najibullah Khairkhwah, 33. "Both can shoot you, rob you. So I prefer not to leave my house."

Big bets aim to jolt heart drug sales back to life

MUNICH (Reuters) - The expiry of patents on profitable heart medicines is soon going to erode one of the main sources of drug company profits, and risky bets on new medicines to replace them may not make up for the lost sales.

High-octane salesmanship was still on show at giant trade stands during this week's European Society of Cardiology congress - the world's biggest heart meeting with 28,000 participants - but the razzmatazz hides a business in trouble.

Patent expiries mean annual sales of 15 different categories of heart drugs are set to fall by more than a quarter by 2017, from $83 billion in 2011 to $60 billion, according to consensus analyst forecasts compiled by Thomson Reuters Pharma.

This year will see the biggest single hit, after the loss of patent protection on Pfizer's record-selling cholesterol pill Lipitor and Sanofi and Bristol-Myers Squibb's blood thinner Plavix.

Those two medicines were the biggest sellers in any disease area last year, with combined global turnover of $20 billion - a figure likely to be halved in 2012.

Big Pharma hopes a new generation of drugs can replace them, yet recent product launches have been sluggish and new heart medicines still in tests represent some of the biggest gambles ever made in the industry's molecular casino.

Leading cardiologists, who follow drug development closely, warn that the hurdle for pricey new heart medicines is getting ever higher as economic pressures hit health budgets worldwide.

"We have a lot of good generic drugs out there and I think doctors are more cognizant of the economic consequences of the therapies they are prescribing than ever before," said Douglas Weaver, head of cardiology at Henry Ford Health System in Detroit.

With new mass-market heart drugs requiring huge clinical trial programs costing up to a $1 billion to prove both safety and efficacy, these are anxious times for the industry.

"It must be making the CEOs of the companies pretty nervous," said Weaver.

"GOOD" CHOLESTEROL

Deutsche Bank analysts believe some new drugs could, in theory, generate annual sales of $10 billion, since there is still plenty to play for in reducing the rate of heart attacks and strokes.

But the risks are high, raising questions for shareholders as to whether their money is being well spent.

Merck & Co, for example, announced this week that more than 20,000 patients had been enrolled into a trial of anacetrapib, a drug to raise levels of "good" HDL cholesterol, which would in theory reduce the risk of heart disease.

However, two similar drugs from Pfizer and Roche have already failed, leaving doctors wary about the approach.

"I think we are a long way from showing that raising HDL really works. I've haven't seen anything that convinces me that we have a winner yet," said Russell Luepker, a heart expert at the University of Minnesota.

Merck is not alone in betting big. Sanofi and its partner Regeneron Pharmaceuticals last month launched a 22,000-patient program to test a new kind of cholesterol fighter that blocks a protein called PCSK9.

Heinz Drexel, a cardiologist at the Vorarlberg Institute for Vascular Investigation and Treatment in Feldkirch, Austria, said it was a "fascinating" approach, but he questioned whether the need to inject the drug every two weeks would curb its use.

GlaxoSmithKline, meanwhile, is testing a heart drug called darapladib in trials involving 27,000 patients, despite mixed results with the compound in mid-stage clinical tests.

GSK gained full control of the product, along with lupus drug Benlysta, after agreeing to buy U.S. biotech firm Human Genome Sciences last month.

Even if they get to market, recent experience suggests these products may have a tough time carving out sales.

Both Eli Lilly and AstraZeneca have struggled to get newer and more potent rivals to Plavix adopted, and Lilly suffered a fresh setback this week when its drug Effient failed to beat Plavix in a key trial.

REPLACING WARFARIN

One area where drug companies and industry analysts do see a big market emerging is in oral anti-coagulants offering an alternative to Warfarin, a notoriously difficult drug to use. Anti-coagulants are used to prevent diseases such as thrombosis, caused by blood clots.

But even here sales expectations have fallen substantially in the past two years as take-up has disappointed.

Data two weeks ago from a U.S. patient registry found only 12.6 percent of those eligible were being prescribed Boehringer Ingelheim's Pradaxa or Bayer and Johnson & Johnson's Xarelto - the first two new anti-coagulants on the market - amid concerns over price and possible bleeding risks.

The picture of slow take-up is apparently similar in Europe. Drexel at the Vorarlberg Institute reported use of the new agents in western Austria was also around 13 percent.

Steen Dalby Kristensen, consultant cardiologist at Denmark's Aarhus University Hospital, said: "We will take it gradually and try and get more experience with the drugs."

"The trend we see with a lot of pharma companies is that they are more and more focusing on specialty markets and selectively exiting the primary care space or downsizing their exposure," she said.

"It doesn't make sense to maintain the large sales forces unless they have a compelling late-stage pipeline."

Two opposing teams battle it out to regulate gene expression on the DNA playing field. One, the activators, keeps DNA open to enzymes that transcribe DNA into RNA. Their repressor opponents antagonize that effort by twisting DNA into an inaccessible coil around histone proteins, an amalgam called chromatin, effectively blocking access to DNA by enzymes that elongate an RNA strand.

Both teams maneuver by chemically modifying histones?the activators by decorating histones with acetyl groups?let's call them green flags?causing them to loosen their grip on DNA. The repressors retaliate by marking histones with red flags, often methyl groups, which call in de-acetylase enzymes to clip off the green flags, restore the chromatin barrier and end that round of gene expression. Disturbing this biochemical balance lies at the heart of many diseases, particularly cancer.

Recently, the lab of Jerry Workman, Ph.D., investigator at the Stowers Institute for Medical Research, reported in the journal Nature that a reserve of "pre-acetylated" histones sits on the chromatin sidelines ready to sub for histones whose green flags get clipped by repressors, a tactic aiding the activators called "histone exchange". In a companion study published in the Aug. 26, 2012 Advance Online Publication of Nature Structural & Molecular Biology the Workman lab now shows that a repressor called Set2 in yeast recruits a protein assistant to block the histone exchange. That study reveals a heretofore unknown mechanism to keep gene activation under control and ensure that erroneous transcription does not occur.

"Accurate gene expression is critical for normal cell function, and when this control is lost cells grow abnormally," says Workman. "These two studies identify mechanisms used by cells to regulate gene expression, which is important for our understanding of what goes wrong in diseases marked by unregulated cell growth, like cancer."

The study began when the group, in collaboration with Stowers proteomics experts Michael Washburn, Ph.D., and Laurence Florens, Ph.D., applied mass spectrometry analysis to identify any protein expressed in yeast Saccharomyces cerevisiae that bound to chromatin in regions patrolled by Set2. Those regions were readily apparent by the presence of Set2's red flag methyl group planted in a specific histone protein interacting with DNA.

"We knew that Set2 added this mark in the middle and downstream parts of genes to recruit de-acetylases," says the study's lead author Michaela Smolle, Ph.D., a postdoctoral researcher in the Workman lab. "But the proteomic search allowed us to cast a wide net for other proteins associated with that mark?a bit like fishing."

Among the fish caught was a component of a yeast chromatin "remodeler" known as Isw1, providing circumstantial evidence that the Set2 red flag attracts Isw1 as well as de-acetylases. Additional genomic experiments evaluating the entire genome of a yeast mutant lacking Set2 supported that idea: not only were the red methyl flags missing but the chromatin landscape was devoid of Isw1 as well.

To assess Isw1's biological function the group exploited yet another yeast mutant, this one lacking the ISW1 gene itself. Microarray analysis of global transcription in ISW1 CHD1 mutants showed widely perturbed gene expression marked by aberrant expression of RNA snippets rather than complete transcripts. Biologists view the presence of such "cryptic transcripts" as indicators of cellular stress.

Analysis of acetylation and methylation patterns in chromatin of ISW1 mutants revealed the probable cause: mutants showed ramped up histone exchange activity marked by excessive levels green-flagged pre-acetylated histones along the length of many genes, a condition likely favoring initiation of truncated RNAs.

"Our work shows that the Set2 methylation mark plays two important roles to ensure that RNA transcription starts only at the beginning of the gene and not in the middle," says Workman. "On one hand, it recruits Isw1 to block incoming histones, and on the other it also recruits a deacetylase to remove any acetylation marks that might happen to have sneaked in."

The Workman lab uses yeast, fruit flies and mammalian cells to study multiple factors that activate and repress transcription. Thus far, they have characterized players in the repressive Set2 pathway primarily in yeast, in part because yeast represents an ideal organism for mutational analysis and has a fairly small genome?6,000 genes compared to the 25,000 or so in humans or mice.

Smolle notes that although yeast genomes are simpler, the principles that govern Set2 activity in yeast will likely hold true for its human counterparts, among them the human protein SETD2. "Yeast has a single methylase, while humans have several, and flies and humans have several Isw1-like proteins," she says. "While you cannot equate one with the other precisely, you can be reasonably sure that what happens in yeast happens in humans as well but tends to be more complicated."

Workman agrees, adding that these discoveries have significant implications for human disease. "Strikingly, the human homolog of Set2, SETD2, is implicated as a tumor suppressor in breast cancer and in renal carcinoma, and those cells are deficient in the SETD2 methylation mark," he says. "These findings strongly suggest that SETD2 could be important in controlling cell growth and preventing tumors. Thanks to yeast, we now know more about how Set2 does that."

CAIRO (AP) ? Egypt's Islamist president Mohammed Morsi on Monday named a team of 21 advisers and aides that includes three women and two Christians and a large number of Islamist-leaning figures, backing off campaign promises to appoint a Christian and a woman as vice presidents.

When asked, Morsi's spokesman Yasser Ali told reporters that there will be only one vice president for the time being.

Instead, Ali on Monday announced the formation of Morsi's "presidential team," which includes four senior aides and a 17-member council of advisers, which includes seven figures seen as political liberals and 10 who have Islamist leanings of various degrees.

The rolling back of the promises reflects Morsi's growing confidence as a president who holds "super powers" exceeding those of his predecessors, said Nabil Abdel-Fatah, a scholar with Al Ahram Center for Political and Strategic Studies. Morsi holds both executive power and legislative authority after he sidelined the top military generals who ruled Egypt after Mubarak's ouster on Feb. 11. 2011. The generals had dissolved parliament and taken on legislative powers, so when they were sidelined, Morsi seized the power to make laws ? a power he has used once so far.

"The announcement of the new team has nothing to do with the promises Morsi made before," said Abdel-Fatah. "Those chose will pose no challenge to the president ... this is only for cosmetic purposes."

Abdel-Fatah said the appointments suggested Morsi does not want to share powers with a vice president. "This is just another sign that we are heading to a deadlock with the Brotherhood insisting on monopolizing power," he said.

The four senior aides include Pakinam el-Sharkawi, a female political scientist who will be in charge of "political issues" and Coptic Christian writer Samer Morcus, whose title is "aide for democratic transition." The oterhs are a senior Brotherhood member, Essam Hadad, who will be the aide for foreign relations and international cooperation and a leading Salafi, Emad Abdel-Ghafour, who was named the president's aide for "society outreach."

The 17-member council includes two women and a Christian, Rafiq Habib, who is deputy leader of the Muslim Brotherhood's political arm Freedom and Justice party and one of the few Christian members of the Islamist group.

The council includes six members of the Muslim Brotherhood's Guidance Bureau, the group's top executive body, and three other figures seen as Islamist-leaning.

Ali, the spokesman, did not say what the council's role would be but said it will "meet regularly."

Ahmed Maher, a co-founder of the secular April 6 youth group, which engineered last year's anti-Mubarak uprising and which has thrown its backing behind Morsi, said that is group was watching how much the president keeps his promises "and we will hold him accountable later on when the time comes."

Though their role is still vague, but Maher believes the aides and the advisers can help Morsi reform state institutions and break away from old regime policies.

"They will be his eyes to carry reforms and institutional shake-ups," said Maher, who comes under criticism for siding by Morsi.

The list did not include three figures that Morsi sidelined in the past month and named as advisers ? the former military chief and defense minister, Field Marshal Hussein Tantawi, his deputy Gen. Sami Anan, and former Prime Minister Kamal el-Ganzouri. That could suggest that their advisory appointments were largely symbolic.

Surely all of that requires a JD.

UnitedHealth doesn't think so. They're requiring an undergraduate degree or "equivalent work experience." They also want five or more years experience working in government or healthcare with experience in compliance; two or more years experience with regulatory filings; and two or more years experience performing data analysis and Market Conduct Exams (bet you didn't learn about those in law school). Even more intriguing, this is a telecommuting job advertised as especially attractive for retirees.

3. ?What about this one: BP needs a Coordinator of Intellectual Property?at its Naperville, Illinois complex. The Coordinator will manage the IP portfolio, advise on all IP related issues, develop IP strategies, help negotiate consulting, confidentiality, services and collaboration agreements, coordinate IP training of staff, and so on. ? Once again, BP doesn't think a JD is necessary for this job. Instead, the company is particularly interested in hiring "project analysts or project managers." For educational background, BP wants only a "bachelor's degree in a technical or business discipline." 4. Finally, the Kiewit Company (a major construction and engineering firm) is looking for a District Compliance Manager. This job requires a ?working knowledge of all applicable local, state and federal compliance regulations; to include, but not limited to, Antitrust/Competition, Foreign Corrupt Practices, Small Business/Disadvantaged Business Enterprises, Transportation, Contract Payments and Claims, Safety, Environmental, and all Employment regulations.?

Few JDs would claim a working of knowledge of that many specialized practice areas. That's fine with Kiewit, because they're not looking for a JD. Instead, they're seeking someone with "a?baccalaureate degree in a construction-related field or equivalent experience and education and a minimum of 3-8 year?s experience in the compliance field.? You also have to be able to lift and carry heavy items weighing approximately 20 pounds.

Nestle, UnitedHealth, BP, and Kiewit still employ lawyers; so do other businesses. But these businesses do not need nearly as many lawyers as our heavily regulated economy suggests. I draw three lessons from job postings like the ones listed here.

First, law schools need to learn much more about the job market for legal services--including the many ways in which nonlawyers perform those services. Rather than dismiss declining job outcomes as a symptom of the recession or the low point in a cycle, law schools need to explore the profound changes sweeping our economy. Computers, off-shoring, out-sourcing, unbundling, and the assignment of legal tasks to nonlawyers: these are real forces, not figments of a few fertile imaginations.

Second, law schools need to rethink the type and cost of the education they offer. We have been producing the same product for at least thirty years. We've updated the look and improved a few features, but legal education in 2012 is not very different from legal education in 1982--except that it costs much, much more. The jobs described above tell us that many consumers are no longer interested in our product: They have found cheaper, better ways to obtain legal services. We can continue to ignore that part of our market, producing only the type of legal service providers we want to produce, but consumers are going to continue falling away.

Finally, these jobs cast doubt on the easy assertions many law schools make about "alternative careers." I found these jobs because I was researching some of those careers. When companies advertise for compliance officers, contract managers, and IP managers, do they show any preference for JDs? I hope there are some who do, but I did not find evidence of that in the postings I read. ?Instead, all of the postings I saw resembled the ones above: The employers wanted very specific workplace experience handling legal documents, rules, and issues of interest to them. I have no sense that any of the above employers would pick up a resume from a recent law graduate and say "What was I thinking? We need a real lawyer in here. This one doesn't have any practical experience but she's from a top tier school and competed in moot court. I'm sure she'll learn all the other stuff on the job."

Jennifer Aniston and Justin Theroux recently announced their engagement, and while he bride-to-be would love to marry abroad, the groom is hoping for their nuptials to take place close to home.

A source claimed the former Friends actress wants a "destination wedding, kind of like eloping with a few friends. There will be the feeling of a vacation, since that's their favourite thing to do."

However actor/screenwriter Justin has other ideas. The insider told website UsMagazine.com: "Justin wants a New York City winter wedding."

The tone and theme of their future nuptials is also a source of contention. Although 43-year-old Jennifer will want to "celebrate in high style," the friend explained: "Justin isn't as flashy as the men she's been with in the past, so it won't go too over-the-top."

Lilly Alzheimer drug slows decline in some; shares jump

NEW YORK (Reuters) - Eli Lilly and Co said its experimental Alzheimer's drug slowed cognitive decline in patients with a mild form of the disease, though it failed key trial goals, sending shares up as much as 8 percent on hopes the treatment may be salvaged.

The late-stage trials, known as EXPEDITION 1 and 2, tested solanezumab in patients with mild-to-moderate Alzheimer's disease, compared with a placebo.

A secondary analysis of the results showed a statistically significant slowing of cognitive decline in patients with mild Alzheimer's disease, but not in those with moderate Alzheimer's, the company said on Friday.

"We recognize that the solanezumab studies did not meet their primary endpoints, but we are encouraged by the pooled data that appear to show a slowing of cognitive decline," said Lilly Chief Executive John Lechleiter. "We intend to discuss these data with regulatory authorities to gain their insights on potential next steps."

Lilly shares climbed 4 percent to $44.10 in early trading on the New York Stock Exchange, after earlier gaining as much as 8 percent.

The results bolstered hopes the treatment may still prove viable for patients either at an earlier stage of the disease, or who have yet to show symptoms.

"Basically the odds of approval just went up from like 10 percent to maybe 20 to 30 percent," said Mark Schoenebaum, analyst with ISI Group, noting that patients with a mild form of the disease comprise at least half of the diagnosed population. "(That's) still high risk, but much better than expected."

Lilly's drug aims to reduce a toxic protein found in the brains of Alzheimer's patients, known as beta amyloid.

Pfizer Inc and Johnson & Johnson earlier this month said their similar drug, bapineuzumab, failed in late-stage trials to improve memory or thinking skills in patients with mild and moderate forms of the disease.

Sanford Bernstein analyst Tim Anderson said the Lilly results on Friday supported a theory that such treatments "probably need to be given as early as possible in the course of the disease."

"At this point, theoretically all possibilities are still on the table," Anderson wrote in a research note. "This includes the possibility that solanezumab could be filed for regulatory approval, given the large unmet medical need in patients with Alzheimer's disease and the generally acceptable safety profile."

"Our best guess is that more studies would have to be done before FDA or other regulatory agencies would consider approving solanezumab," he said.

Alzheimer's is the most common form of dementia and the sixth leading cause of death in the United States. An estimated 5 million Americans are believed to have the disease.

The Lilly trials tested solanezumab in more than 2,050 patients in 16 countries, over an 18-month period. Patients on the drug also experienced adverse events such as lethargy, rash, malaise and angina, compared to the placebo.

The company said more detailed data will be presented at medical meetings in Boston and Monaco in October.
